About the ABI PRISMĘ BigDye(tm) Chemistry V3 Updater ---------------------------------------- This document pertains to the CD labeled: ABI PRISM Ę BigDye(tm) v3 Chemistries Software Support Files PN 4326478, For Windows NTĘ platform ---------------------------------------- This file contains the description of software support required to analyze sequencing data generated with ABI PRISMĘ BigDye(tm) v3 chemistry on the following instruments: 1. ABI PRISMĘ 3700 DNA Analyzers 2. ABI PRISMĘ 3100 Genetic Analyzers 3. ABI PRISMĘ 377 DNA Sequencers (Mac and NT platforms) 4. ABI PRISMĘ 310 Genetic Analyzers (Mac and NT platforms) 5. ABI PRISMĘ 373 DNA Sequencers (Mac and NT platforms) This media is compatible on computers running Windows NTĘ operating system. The new ABI PRISMĘ BigDye(tm) v3 chemistry includes: - BigDye(tm) v3 Terminator - BigDye(tm) v3 Primer Forward (-21 M13) - BigDye(tm) v3 Primer Reverse (-21 M13) - Long Read Standard for making matrices on 3100 and 3700 instruments The purpose of this updater is to provide all the necessary software to perform sequencing applications using BigDye(tm) v3 chemistry on existing instruments in the field and new instruments for all the genetic analyzer instruments listed above. The BigDye(tm) v3 chemistry does not affect 5th dye Lane Guide(tm) applications or GeneScanĘ applications. HOW TO USE THIS UPDATER ----------------------- Ensure that you have installed Sequencing Analysis software or 3700/3100 DataCollection software on your computer. If you have installed into: D:\AppliedBio, or D:\Perkin-Elmer, the Updater will automatically find that folder and install the files into their appropriate places. If these files already exist (same versions), they will not be replaced. 1. a. Double-click the My Computer icon. b. Double-click the CD-ROM icon. c. Double-click the Setup.exe icon. 2. Click Next to go through the first few dialog boxes. 3. In the Select Instrument dialog box, select the appropriate ABI PrismĘ instrument(s), then click Next. 4. In the Ready To Install dialog box, click Next. Installation starts by copying the necessary support files. 5. When the installation is finished, click Finish to terminate the program. *Note--Sequencing Analysis must be restarted in order to see the new DyeSet/Primer files in the Sample Manager. ************************************** YOUR SOFTWARE IS NOT LOCATED IN D:\AppliedBio folder or D:\Perkin-Elmer folder? On analysis computer, not attached to an genetic analyzer instrument, if you installed Sequencing Analysis Software into a folder other than D:\AppliedBio or D:\Perkin-Elmer, the installer will NOT automatically install the files. To manually install the files needed to analyze sequencing data generated with ABI PRISMĘ BigDye(tm) v3 chemistry do the following: 1. a. Search on your system for the location where you installed Sequencing Analysis software. b. Locate the folder: ...\Shared\Analysis\Basecaller\Mobility\ 2. a. Double-click the My Computer icon. b. Double-click the CD-ROM icon. c. Double-click the folder labelled "BigDyeV3Chemistry_SoftwareSupportFiles" d. Locate the folder with the instrument number that you have (3700, 3100, 373&377 or 310) e. Copy the CONTENTS of the folder into your ...\Shared\Analysis\Basecaller\Mobility\ folder. Do NOT copy then entire folder, just copy the CONTENTS. f. If you need to analyze data from more than one type of instrument then repeat step D. *Note--Sequencing Analysis must be restarted in order to see the new DyeSet/Primer files in the Sample Manager. ************************************** What Gets Installed ------------------- This Updater installs new DyeSet/Primer files and (for 3700/3100 instruments) a new spectral calibration parameter file. Different files are installed depending on which instrument is selected during the installation process.
